Ba-Wei-Die-Huang-Wan can Ameliorate Ketamine-induced Ulcerative Cystitis in Rats

Purpose: We investigate the effect of Ba-Wei-Die-Huang-Wan on ketamine-induced ulcerative cystitis in rat’s bladder.
 
Materials and methods: Female Sprague-Dawley rats were subject to three groups, which receive saline, ketamine (25mg/kg/day, i.p.) or ketamine combined with Ba-Wei-Die-Huang-Wan (90mg/kg/day) treatment. Voiding behavior of rats was evaluated by metabolic cage study, and cystometry. Paraffin-embedded sections were stained. Bladder mucosa and muscle proteins were also assessed by Western blotting.
 
Results: As compared to controls, the ketamine group showed increased urinary frequency, and bladder overactivity, whereas the Ba-Wei-Die-Huang-Wan group did not. The ketamine group had significant protein overexpression in bladder mucosal TRPV1 receptor, and detrusor M2, M3 receptors. Also, the ketamine group had significantly increased inflammatory markers, including TNF-α, and ICAM-1 in detrusor. However, the ketamine/Ba-Wei-Die-Huang-Wan group had insignificant changes from controls.
 
Conclusion: Ba-Wei-Die-Huang-Wan intake can ameliorate bladder overactivity and hypersensitivity of ketamine-induced ulcerative cystitis of rats by decreasing bladder inflammatory signals of TNF-α, and ICAM-1, and suppressing mucosal TRPV1 receptor as well as detrusor M2 receptors overexpression.
