Lactobacillus plantarum reforms reproductive function in male mice with high-fat diet-induced obesity

Purpose:

This study investigated the effects of Lactobacillus plantarum 1008 (LP1008) supplementation on metabolic dysfunction, testicular antioxidant capacity, apoptosis, autophagy, and spermatogenesis in male mice with high-fat diet-induced obesity.

Materials and Methods:

Thirty-six male C57BL/6 mice were fed a normal diet (NC group, n =12) or a high-fat control diet (fed with 45% kcal from fat, HFC group, n = 24) for 16 weeks, and mice randomly chosen from the HFC group were later treated with a probiotic (LP1008) for the final 8 weeks of HFP feeding (high-fat diet+LP1008, n = 12).

Results:

The results showed that compared with the HFC group, the obese mice supplemented with the probiotic (HFP group) exhibited improved blood cholesterol, insulin resistance, AST, ALT, and lipopolysaccharides (LPS). The HFC group exhibited a lower testicular testosterone level and sperm quality and a lower 17β-HSD protein expression, which were improved by LP1008 treatment. On the other hand, the HFC group had lower SOD, catalase, and glutathione peroxidase enzyme activities, and LP1008 treatment increased the antioxidative activities and reduced lipid peroxidation. The HFC group also exhibited increased apoptosis, inflammation, and autophagy proteins in the testis, which were improved by LP1008 supplementation. Moreover, the gut microbiota analysis results showed that the Firmicutes/Bacteroidetes ratio was significantly increased in the HFC and HFP groups compared with the NC group, and LP1008 treatment decreased Ruminococcaceae and increased Bifidobacteriaceae diversity.

Conclusion:

LP1008 treatment increased antioxidative enzyme levels and regulated microbiota-ameliorated HFC-induced oxidative stress, apoptosis, inflammation, and autophagy and thus reformed testicular function and semen quality.
