Alteration of cannabinoid receptor by phlorizin in type I diabetic rats’s bladder

Purpose: To investigate Alteration of cannabinoid receptor (CB) by phlorizin in type I diabetic rats’s bladder.
Methods: Type 1 DM rats were induced by streptozotocin (STZ) for 2 weeks. Three kinds of treatment was applied: 1. Insulin; 2. Phlorizin (a sodium glucose co-transporter inhibitor,); and 3. Vehicle. In an organ bath, bladder muscle strips, including normal rats, were prepared with 1 μmol/L acetylcholine (ACh). CB1/CB2 agonist was added and the response of muscle strip to the agnosit was examined. Blotting and real-time polymerase chain reaction (RT-PCR) analyses were used to identify CB1/CB2 receptor protein and mRNA expressions.
Results: High blood glucose level was found in type 1 diabetic rats. Insulin and phlorizin treatment significantly reduced the elevated glucose level. Less relaxation response developed in the type 1 diabetic rats and CB1/CB2 relaxation effect improved after insulin and phlorizin treatment. CB1/CB2 receptors protein and mRNA level were reduced in the diabetic rats’ bladder. The receptors expressions became normal after insulin and phlorizin treatment.
Conclusions: Decreased CB1/CB2 receptors levels were found in type 1 diabetic rats’ bladder and the receptors expressions became normal after phlorizin treatment.
